Why Free Form L-Tryptophan

A single amino acid with a detailed impurity specification

L-tryptophan has one of the most detailed specifications of any amino acid. Beyond assay and rotation, good specs name trace byproducts and set HPLC limits for them. That gives quality teams more to check, and more to compare between lots.

One named compound

L-tryptophan has the formula C11H12N2O2 and a molecular weight of 204.23. It is sold without a salt partner, so a 99% assay means about 99% of the powder is tryptophan.

Optical identity you can check

Specific rotation confirms the L-form. Published specifications call for about −29.4° to −32.8°, and a racemic DL material would read near zero.

Named byproducts with limits

Pharmacopeia specs set HPLC limits for named trace byproducts. One widely cited example is EBT, 1,1'-ethylidenebis(L-tryptophan), which the European Pharmacopoeia limits to NMT 10 ppm.

Fermentation source

Commercial L-tryptophan is made by microbial fermentation, then purified with steps such as activated carbon and crystallization. Each step belongs in the supplier's process record.

White crystals that need shade

Good lots are white crystals or crystalline powder. Published storage notes call for protection from heat, light, and oxygen, so keep it in opaque packaging.

Trade rules to check

21 CFR 172.320 lists L-tryptophan for US conventional foods at up to 1.6% of total protein. Rules in other countries differ, and several restrict its sale.

Formulation Uses

Where formulators use L-tryptophan powder

L-tryptophan fits measured formats where fill weight and serving size are controlled, including:

  • Capsules
  • Compressed tablets
  • Single ingredient powders
  • Multiple ingredient amino acid blends
  • Stick packs and sachets
  • Powdered drink mixes, after taste trials
  • Private label programs

Plan fill weights from the confirmed assay on each COA. Check the rules in every market where the finished product will be sold before you lock a formula, because tryptophan rules differ by country.

What Commercial Buyers Should Review

What to check before you buy L-tryptophan

Powders sold as tryptophan can differ in impurity profile, process controls, and the markets they can legally enter. Review these points together, and ask for lot documents before you approve a supplier.

Name and CAS

Confirm that the specification and certificate of analysis (COA) both name L-tryptophan, CAS 73-22-3. 5-HTP is a different compound, usually made from Griffonia simplicifolia seed.

HPLC related compounds

Ask for related compounds by HPLC, with EBT reported by name. A Ph. Eur. grade lot should show EBT at NMT 10 ppm.

Process change control

Ask the maker to name its production strain and purification steps. Then ask for written notice before any change to either.

Assay and rotation

A typical assay limit is 98.5 to 101.5% on a dried basis, with rotation near −29.4° to −32.8°. Check both on every COA.

Contaminant limits

Check lead, arsenic, cadmium, and mercury by ICP-MS, plus microbial limits by USP methods. Compare each limit with your own finished product specification.

Destination market rules

Tell us where the finished product will be sold. Some countries restrict tryptophan, so export plans should be checked before you order.

Label math: A 500 mg serving at a 99.0% assay holds about 495 mg of L-tryptophan. Base label figures on each lot's measured assay and your own finished product testing, since the stated grade is only a minimum.

Specification

Typical specification for L-tryptophan

These values reflect published USP and Ph. Eur. grade specifications. Final figures come from the specification sheet and COA for your lot. NLT means not less than, and NMT means not more than.

Storage & Handling

Storage and handling for bulk L-tryptophan

L-tryptophan keeps best when it stays sealed, dry, and out of light. These steps keep the powder in good condition between production runs.

Keep it sealed

Close the inner liner after each use and press out the air. Less air in the bag means less contact with oxygen and moisture.

Keep away from light

Leave the powder in its original opaque packaging. Published storage notes call for protection from light, and color can shift with long exposure.

Store cool and dry

Keep bags off the floor in a dry room, away from heat sources. Follow the temperature range on the spec sheet.

Rotate stock

Record the lot number and retest date for each bag. Send older lots to production first.

Buyer FAQ

L-tryptophan buyer FAQ

Quick answers to the questions procurement and quality teams ask most.

Why do L-tryptophan specifications list named impurities?

Trace fermentation byproducts are a known concern for this amino acid. After a 1989 contamination case was traced to one producer's process changes, pharmacopeia specs added HPLC limits for named byproducts. Good lots report them by name on the COA.

What is EBT in L-tryptophan?

EBT is 1,1'-ethylidenebis(L-tryptophan), a trace byproduct measured by HPLC. The European Pharmacopoeia limits it to NMT 10 ppm.

Is L-tryptophan the same as 5-HTP?

No. 5-HTP (5-hydroxytryptophan) is a different compound, usually made from Griffonia simplicifolia seed. It has its own specification and its own product page.

What specific rotation should L-tryptophan have?

Published specifications call for about −29.4° to −32.8°. A result inside that range confirms the L-form, while a racemic DL material would read near zero.

Can L-tryptophan be sold in every country?

No. In the US it is sold as a dietary ingredient and is listed for conventional foods under 21 CFR 172.320. Several other countries restrict it, so check each export market before you quote.

How is L-tryptophan made?

L-tryptophan is made by microbial fermentation, then purified and crystallized. Ask your supplier to name the strain and purification steps, and to notify you of any change.

What documents can I get with an order?

A certificate of analysis (COA) and a specification sheet ship with every order. An allergen statement and a safety data sheet may also be available, depending on the lot in inventory. Contact us to request them.

Can I get a sample before a bulk order?

You can request one by choosing sample options on the quote form. We will confirm availability, size, and timing.
