Bulk 5-HTP Powder from Griffonia Seed for Supplement Manufacturing

5-HTP is a purified amino acid powder made from the seeds of Griffonia simplicifolia, a West and Central African shrub in the legume family. Its full name is L-5-hydroxytryptophan, and purity is measured by HPLC. We supply it in bulk to supplement brands, contract manufacturers, and formulators. You can request testing documents and samples on the quote form.

Why 5-HTP Powder

A high purity seed extract that is easy to specify

Bulk 5-HTP is sold at 98% to 99% purity, so one HPLC result describes nearly all of each lot. A named plant source and standard USP tests make supplier comparison simple for buyers and quality teams.

One compound, one number

Most grades are 98% to 99% L-5-hydroxytryptophan by HPLC. At that purity, fill weight and label amount are nearly the same, which keeps your formula math simple.

Seed from West and Central Africa

Griffonia simplicifolia is a woody shrub that grows in West and Central Africa, with cultivation in Ghana, Ivory Coast, and Togo. Published seed analyses report about 10% to 20% 5-HTP by weight.

Extracted, then crystallized

Producers extract the ground seed, then purify the 5-HTP into a white to off white crystalline powder. Ask your supplier to name the extraction solvent and send the residual solvent result.

HPLC is the reference method

5-HTP, tryptophan, and related indole compounds give strong UV readings in the same range, between 230 and 300 nm. HPLC separates them before measuring, so it is the method to ask for on every COA.

The L-form, checked by rotation

Seed derived 5-HTP is the L-isomer, registered as CAS 4350-09-8. A specific optical rotation test confirms the form, and spec sheets give a narrow range for it.

Fine, dense crystals

Typical lots pass an 80 mesh screen, with bulk density at or above 0.38 g/mL and tapped density at or above 0.58 g/mL. Small fill weights fit easily into small capsules and tablets.

Formulation Uses

Where formulators use 5-HTP powder

This powder fits measured formats where fill weight per unit is controlled, including:

  • Capsules
  • Compressed tablets
  • Chewable tablets, after taste trials
  • Single ingredient powders for encapsulation
  • Multiple ingredient capsule and tablet blends
  • Gummies, after taste and stability trials
  • Private label programs

Plan fill weights from the HPLC result on each COA, and note whether it is on an as is or dry basis. Small label amounts often need a diluent or flow agent to reach a fill weight your machine can hold steady. Run a blend uniformity test before you lock in capsule weights.

What Commercial Buyers Should Review

What to check before you buy 5-HTP powder

Two powders sold as 99% 5-HTP can differ in source, assay method, and impurity profile. Review these points together, and ask for lot documents before you approve a supplier.

Source declaration

Confirm that the specification names Griffonia simplicifolia seed as the only source of the 5-HTP. It can also be made by microbial production, so ask for a written declaration if your label says it comes from the seed.

Purity method and basis

Ask for purity by HPLC against a reference standard, and whether the result is on an as is or dry basis. A UV result cannot tell 5-HTP apart from related indoles in the same sample.

Impurity profile

Ask for total related substances by HPLC, often set at NMT 0.5%. Then ask whether the COA also reports named peaks, such as peak E and 4,5-tryptophan-dione, by HPLC or LC-MS.

Seed origin and processing site

Ask for the country where the seed was harvested and the country where it was extracted. Supplier listings name Ghana, Nigeria, and Togo for seed, and India for extraction.

Carrier and additives

A 98% or 99% grade should contain no added carrier. Confirm that the specification says none, and ask for an allergen statement for the lot.

Contaminant limits

Check heavy metals by ICP-MS, microbial limits by USP methods, and residual solvents against USP <467>. Compare each limit with your own finished product specification.

Label math: If your label lists 5-HTP per capsule, base the number on each lot's measured purity and your own finished product testing. A stated grade is a minimum, so the measured value on the COA is the figure to work from.

Specification

Typical specification for 5-HTP powder

These values reflect the common 99% HPLC grade. Final figures come from the specification sheet and COA for your lot. NLT means not less than, and NMT means not more than.

Storage & Handling

Storage and handling for bulk 5-HTP

5-HTP holds its quality longest when it stays sealed, dry, and out of light. These steps keep the powder in good condition between production runs.

Keep it sealed

Close the inner liner after each use and press out the air. Air and moisture speed oxidation, and an oxidation product of 5-HTP is one of the peaks labs track.

Keep away from light

Leave the powder in its original opaque drum or bag. Light and heat can shift color and the impurity profile over time.

Store cool and dry

Keep drums off the floor in a dry room, away from heat sources. Follow the temperature range on the spec sheet.

Rotate stock

Record the lot number and retest date for each drum. Send older lots to production first, and retest purity on any lot held past its retest date.

Buyer FAQ

5-HTP powder buyer FAQ

Quick answers to the questions procurement and quality teams ask most.

What is 5-HTP powder made from?

Most bulk 5-HTP is extracted from the seeds of Griffonia simplicifolia, a shrub of West and Central Africa. The extract is purified to a white to off white powder of 98% to 99% L-5-hydroxytryptophan.

Should I choose 98% or 99% 5-HTP?

Either grade works when the method behind it is sound, and 99% by HPLC is common on technical spec sheets. Compare HPLC results on the same basis, then choose on price, impurity profile, and documents.

How is 5-HTP purity measured?

Purity is measured by HPLC against a 5-HTP reference standard. UV readings alone cannot separate 5-HTP from tryptophan and related indoles. LC-MS adds a further check on named impurity peaks.

What impurities should a 5-HTP specification cover?

It should cover total related substances by HPLC, commonly NMT 0.5%, plus residual solvents, heavy metals, and microbial limits. Also ask whether the supplier's HPLC or LC-MS method reports peak E and 4,5-tryptophan-dione by name.

Is 5-HTP the same as L-tryptophan?

No. 5-HTP is L-tryptophan with an added hydroxyl group at position 5, so the two are different compounds. 5-HTP is CAS 4350-09-8, and L-tryptophan is CAS 73-22-3.

What documents can I get with an order?

A certificate of analysis (COA) and a specification sheet ship with every order. An allergen statement and a safety data sheet may also be available, depending on the lot in inventory. Contact us to request them.

Can I get a sample before a bulk order?

You can request one by choosing sample options on the quote form. We will confirm availability, size, and timing.

How should I store an opened drum?

Close the liner, press out the air, and keep the drum in a cool, dry place away from light. Retest purity on any lot held past its retest date.
